Rupali is best known for co-starring with Ratna Pathak Shah in the hit comedy TV series Sarabhai vs. Sarabhai as the middle-class housewife Monisha Sarabhai. Rupali has appeared in a number of television programmes, including Baa Bahoo Aur Baby and Parvarrish - Kuchh Khattee Kuchh Meethi. In her fictional show Anupamaa, in which she portrays the lead role of a Gujarati housewife, she has been dominating the charts for the last few years.
The television actor Rupali Ganguly has admitted that she did not nurse her son Rudransh Verma after his birth and that the social pressure to do so made her feel like a "failed mother." Two years after she wed businessman Ashwin K. Verma in 2013, Rupali gave birth to her son in 2015.

Because of the challenges she encountered before to becoming pregnant, Rupali has frequently referred to her baby as a "miracle." She struggled to get pregnant due to thyroid problems. Rupali added that at the moment, having a child was her only priority in life.

Rupali recalled the moment she gave birth to her son in an interview, "Once the child is born, we women put too much pressure on ourselves that only breast milk should be given, again the conditioning that you have got as a child, you follow that. People come to give you advice. I did not lactate and it is fine. I want to tell all the mothers. I totally killed and judged myself, I felt like a failed mother over it that I can’t even feed my baby. I psyched myself completely."

She continued by stating that she gained weight after consuming various foods to aid in lactation and that she now weighed 83 kg. “After a point even my ankles couldn’t bear my weight. When I would take my son to the pram for a stroll, people would say 'Arey Tu toh Monisha hai Na, kitni Moti hogayi'(You are Monisha! You have become fat).' Maybe someone must have said that in a very nice way, but you feel bad.” Such remarks, according to Rupali, do hurt. She also recalled being told she had changed into a "aunty" by a friend who is also an actress.
